Pexidartinib’s Midazolam Interaction Differed After Single and Repeated Exposure

Written by

in

An interaction cannot always be summarized by one time-independent percentage.

What the supplied paper shows

Clinical probe studies evaluated pexidartinib with substrates of several enzymes and transporters. Midazolam exposure decreased more after multiple pexidartinib doses than after a single dose, supporting a treatment-duration-dependent CYP3A effect.

Limits of the finding

The study regimens and selected participants limit individual predictions. A group pharmacokinetic change is not an impairment estimate, and effects on other probe drugs did not all follow the midazolam pattern.

What matters in a forensic review

Record the start date, repeat dosing and any discontinuation of the interacting medicine. Do not assume that a first-day interaction describes established therapy. Forensic reconstruction should distinguish modeled or average exposure changes from measured patient concentrations and actual clinical response.
